「もう一度言ってください」「何とおっしゃいましたか？」という意味になっています。
「本当かい？」「うそじゃないだろうね」と言う場合も結構多いです。
“Ted and Lisa are getting married!”
「テッドとリサが結婚するのよ！」
“Come again?”
「もう一度言って」
“Ted and Lisa are getting married next month! And they’re only just met!”
「来月テッドとリサが結婚するのよ！それも、まだ出会ったばかりなのに！」
または
“He’s an entomologist.”
「彼は昆虫学者なの」
“Come again, please?”
「もう一度言ってください」
“An entomologist: she studies insects.”
「昆虫学者よ。彼女は昆虫の研究をしているの」
