“I wasn’t here yesterday, can you fill me in?”
“fill”には、「～を満たす；（ページ・空白部分）を埋める」という意味ですが、“fill （人） in”は、「（人の）（情報の空白部分）を埋める」というところから、「（人）に最新の詳しい情報を教える」という意味になります。
“Fill me in.”と言うと、「詳しく説明して」「詳しく教えて」などという意味です。
“fill somebody in on ～”とすると、「～に関して、～について」「（人）に最新の詳しい情報を教える」という意味です。
“Did you hear they’re getting married?”
「彼らが結婚するって聞いた？」
“Really? Fill me in.”
「本当？ 詳しく教えてよ。」
