“Something is fishing, you’re up to something“
“be up to ～”には、「～を企んでいる、（ひそかに）～しようと計画している」という意味です。何かよくないことを企んでいる場合に用いられる表現になります。
“You’re up to something.”とすると、「何か企んでいるでしょ？」という意味です。
“be up to ～”には、単なる予定として「～しようとしている」という意味もあるので、“What are you up to?”とすると、「今から何するの？」という意味、もちろん、上記の意味で、「何を企んでいるんだ？」という意味にもなるので、どちらの意味なのかは、その会話の流れで判断するになります。
“What is it?”
「何なの？」
“Oh, nothing.”
「何でもないよ」
“You’re up to something.”
「何か企んでいるでしょ？」
